You can use the email_response probe to actively test an email system. It sends messages and looks in a mailbox for them to be delivered. My recommendation would be to setup the probe to talk to an email system local to NMS and have it send test messages to an email address in the system you want to monitor. That address can be configured to forward the messages back out to a mailbox on the email system local to NMS, and the probe would be receive the messages and report if any were not returned.
If you are looking for statistics about real email messages flowing through a system rather than to actively test the email system, I do not know of a probe that will do that. I am sure it would depend on the mail system. There is a probe for Exchange, which relies heavily on the Windows performance counters for Exchange. I am not sure if those counters would track this type of information or not.
