Can we create server availability report trrough creating a new probe using Microsoft utility Uptime.exe? So that we can have the availability report exactly same as in SCOM.

You would have to wrap the call to uptime.exe in some language we support. Available options are:
LUA (through the NSA)
Batch files (using utilities)
I guess you would call uptime.exe and parse the output in some fashion. Based on the data you could then generate either/both QoS messages and/or alarms.
Another option I guess would be to run the command in the logmon probe and parse the output there.
I'm not familiar with uptime.exe and what the report contains, so I can't be more specific than this, I'm afraid...
